heal.abstractΗ ανάπτυξη της γονιδιωματικής και της βιοπληροφορικής συνέβαλε στην απαρχή μιας διεξοδικότερης επιστημονικής έρευνας σχετικά με τους μικροοργανισμούς που εντοπίζονται στον ανθρώπινο οργανισμό, το σύνολο των οποίων αποτελεί το ανθρώπινο μικροβίωμα που ενυπάρχει σε διαφορετικά μέρη του σώματος. Η παρούσα διπλωματική μελετά βιβλιογραφικά και πραγματεύεται τη σύνθεση του εντερικού μικροβιώματος, τις μεταβολές του κατά τις διάφορες φάσεις της ζωής του ανθρώπου, καθώς και τους παράγοντες που το επηρεάζουν, με έμφαση στις διατροφικές συνήθειες. Αναλύεται, επίσης, η σχέση του με το πολυπαραγοντικό νόσημα της παχυσαρκίας, μία σύγχρονη πανδημία που μαστίζει την ανθρωπότητα. Αναφέρονται τα αίτια και η ταξινόμησή της, και επιπλέον γίνεται λόγος για την αυξανόμενη πρόκληση της παιδικής παχυσαρκίας. Στη συνέχεια, παρουσιάζεται η σχέση που υπάρχει μεταξύ του εντερικού μικροβιώματος και της παχυσαρκίας και περιγράφονται οι προτεινόμενοι μηχανισμοί με τους οποίους μικροβίωμα επηρεάζει τα θεμελιώδη μονοπάτια του μεταβολισμού και κατ’ επέκταση συμβάλλει στην εξέλιξη της παχυσαρκίας. Επιπροσθέτως, αναφέρονται κάποιες σημαντικές κλινικές δοκιμές και πραγματοποιούμενες έρευνες που επιβεβαιώνουν τα στοιχεία αυτά. Ένα από τα σημαντικότερα ευρήματα αυτής της ανασκόπησης είναι ότι η παθοφυσιολογία της παχυσαρκίας και οι μεταβολικές δυσλειτουργίες που επακολουθούν δεν επηρεάζουν μόνο τον οργανισμό στον οποίο εμφανίζονται, αφού οι διαταραχές της ομοιόστασης της εντερικής χλωρίδας (δυσβίωση) και οι συνέπειες που προκύπτουν, μπορούν να μεταφερθούν μέσω επιγενετικών μηχανισμών από την έγκυο μητέρα στο νεογνό. Επισημαίνεται ακόμα η αξία της διατροφής και συγκεκριμένα της Μεσογειακής Διατροφής, ως κοινή συνιστώσα τόσο για την ισορροπημένη σύσταση και την ομαλή λειτουργία του μικροβιώματος όσο και για την πρόληψη και αντιμετώπιση της παχυσαρκίας, ενώ αναδεικνύεται η ύψιστη σημασία της για την υγεία του οργανισμού συνολικά. Ακολουθούν τέλος τα συμπεράσματα που εξάγονται από την παρούσα βιβλιογραφική μελέτη, καθώς και ορισμένες σκέψεις και προτάσεις για μελλοντική έρευνα.el
heal.abstractThe advent of genomics and bioinformatics has contributed to the initiation of a more thorough scientific investigation of the microbes colonizing the human body which constitutes the human microbiome that resides on different parts of the body. The present diploma thesis deals bibliographically with the composition of the intestinal microbiome, its changes during the various phases of human life, and the factors that influence it with emphasis on eating habits. The relationship of the microbiome with obesity, a chronic, multifactorial disease, a modern pandemic that plagues humanity, is also analyzed. After a thorough discussion of its causes and classification, the growing challenge of childhood obesity is also discussed. Next, the relationship between the gut microbiome and obesity is presented, and the proposed mechanisms by which the microbiome affects fundamental pathways of metabolism and contributes to the development of obesity are described. In addition, some important clinical trials and ongoing research that confirm these facts are mentioned. One of the most important conclusions of this thesis is the pathophysiology of obesity and the metabolic dysfunctions that company it, do not only affect the organism in which they occur, since dysregulation of the gut microbiome (dysbiosis), and the resulting consequences can be transmitted through epigenetic mechanisms from the pregnant mother to the fetus.The value of nutrition, especially the Mediterranean Diet, is highlighted, as a common component both for the balanced composition and the smooth functioning of the microbiome, as well as for the prevention and treatment of obesity. At the same time, its supreme importance for the health of the body as a whole is also emphasized. The conclusions drawn from the present bibliographic study and some thoughts and suggestions for future research are also outlined at the end.en
